Sen. Marco Rubio, R-Fla.. center, answers a reporter's question as he and a bipartisan group of leading senators announce that they have reached agreement on the principles of sweeping legislation to rewrite the nation's immigration laws, during a news conference at the Capitol in Washington, Monday, Jan. 28, 2013. Scott Applewhite)

(AP) ? Declaring "now is the time" to fix the nation's broken immigration system, President Barack Obama on Tuesday outlined broad proposals for putting millions of illegal immigrants on a clear path to citizenship while cracking down on businesses that employ people illegally and tightening security at the borders. He hailed a bipartisan Senate group on a similar track but left unresolved key details that could derail the complex and emotional effort.

Potential Senate roadblocks center on how to structure the avenue to citizenship and on whether legislation would cover same-sex couples ? and that's all before a Senate measure could be debated, approved and sent to the Republican-controlled House where opposition is sure to be stronger.

Obama, who carried Nevada in the November election with heavy Hispanic support, praised the Senate push, saying Congress is showing "a genuine desire to get this done soon." But mindful of previous immigrations efforts that have failed, he warned that the debate would be difficult and vowed to send his own legislation to Capitol Hill if lawmakers don't act quickly.

"The question now is simple," Obama said during a campaign-style event in Las Vegas, one week after being sworn in for a second term in the White House. "Do we have the resolve as a people, as a country, as a government to finally put this issue behind us? I believe that we do."

Shortly after Obama finished speaking, cracks emerged between the White House and the group of eight senators, which put out their proposals one day ahead of the president. Florida Sen. Marco Rubio, a potential 2016 presidential candidate, faulted Obama for not making a citizenship pathway contingent on tighter border security, a central tenet of the lawmakers' proposals.

"The president's speech left the impression that he believes reforming immigration quickly is more important than reforming immigration right," Rubio said in a statement.

House Speaker John Boehner also responded coolly, with spokesman Brendan Buck saying the Ohio Republican hoped the president would be "careful not to drag the debate to the left and ultimately disrupt the difficult work that is ahead in the House and Senate."

Despite possible obstacles to come, the broad agreement between the White House and bipartisan lawmakers in the Senate represents a drastic shift in Washington's willingness to tackle immigration, an issue that has languished for years. Much of that shift is politically motivated, due to the growing influence of Hispanics in presidential and other elections and their overwhelming support for Obama in November.

The separate White House and Senate proposals focus on the same principles: providing a way for most of the estimated 11 million people already in the U.S. illegally to become citizens, strengthening border security, cracking down on employers who hire illegal immigrants and streamlining the legal immigration system.

A consensus around the question of citizenship could help lawmakers clear one major hurdle that has blocked previous immigration efforts. Many Republicans have opposed allowing illegal immigrants to become citizens, saying that would be an unfair reward for people who have broken the law.

Details on how to achieve a pathway to citizenship still could prove to be a major sticking point between the White House and the Senate group.

Obama and the Senate lawmakers all want to require people here illegally to register with the government, pass criminal and national security background checks, pay fees and penalties as well as back taxes and wait until existing immigration backlogs are cleared before getting in line for green cards. Neither proposal backs up those requirements with specifics.

After achieving legal status, U.S. law says people can become citizens after five years.

The Senate proposal says that entire process couldn't start until the borders were fully secure and tracking of people in the U.S. on visas had improved. Those vague requirements would almost certainly make the timeline for achieving citizenship longer than what the White House is proposing.

The president urged lawmakers to avoid making the citizenship pathway so difficult that it would appear out of reach for many illegal immigrants.

"We all agree that these men and women have to earn their way to citizenship," he said. "But for comprehensive immigration reform to work, it must make clear from the outset that there is a pathway to citizenship."

"It won't be a quick process, but it will be a fair process," Obama added.

Another key difference between the White House and Senate proposals is the administration's plan to allow same-sex partners to seek visas under the same rules that govern other family immigration. The Senate principles do not recognize same-sex partners, though Democratic lawmakers have told gay rights groups that they could seek to include that in a final bill.

John McCain of Arizona, who is part of the Senate immigration group, called the issue a "red flag" in an interview Tuesday on "CBS This Morning."

Washington last took up immigration changes in a serious way in 2007, when then-President George W. Bush pressed for an overhaul. The initial efforts had bipartisan support but eventually collapsed in the Senate because of a lack of GOP support.

Cognizant of that failed effort, the White House has readied its own immigration legislation. But officials said Obama will send it to the Hill only if the Senate process stalls.

Most of the recommendations Obama made Tuesday were not new. They were included in the immigration blueprint he released in 2011, but he exerted little political capital to get it passed by Congress, to the disappointment of many Hispanics.

Some of the recommendations in the Senate plan are also pulled from past immigration efforts. The senators involved in formulating the latest proposals, in addition to McCain and Rubio, are Democrats Charles Schumer of New York, Dick Durbin of Illinois, Robert Menendez of New Jersey and Michael Bennet of Colorado, and Republicans Lindsey Graham of South Carolina, and Jeff Flake of Arizona.

Also Tuesday, in another sign of Congress' increased attention to immigration issues, a group of four senators introduced legislation aimed at allowing more high-tech workers into the country, a longtime priority of technology businesses. The bill by Republicans Rubio and Orrin Hatch and Democrats Amy Klobuchar and Chris Coons would increase the number of visas available for high-tech workers, make it easier for them to change jobs once here and for their spouses to work and aim to make it easier for foreigners at U.S. universities to remain here upon graduation.

'Hansel & Gretel' hunts down a box office win

By Reuters

A grown-up "Hansel & Gretel" grabbed the weekend box office title, pulling in $19 million in U.S. and Canadian ticket sales with its reinvention of the classic fairy tale characters as fierce bounty hunters.

"Hansel & Gretel: Witch Hunters" knocked last weekend's winner, low-budget horror flick "Mama," into second place. "Mama" earned $12.8 million from Friday through Sunday, according to studio estimates, followed by "Silver Linings Playbook" with $10 million.

CIA drama "Zero Dark Thirty" came in fourth with $9.8 million.

Jeremy Renner and Gemma Arterton star in "Hansel & Gretel" as crossbow-wielding adult siblings who travel the world to take out evil witches. MGM and Viacom Inc's Paramount Pictures produced the action comedy for about $50 million. Paramount had predicted opening weekend sales in the high-teens or low $20 million range.

Don Harris, Paramount's president of distribution, said the film performed well despite the very cold temperatures and snow that hit the eastern United States.

"We are pleased that we were on our number on a worldwide basis. It looks like we are on or exceeding our numbers, but we did get dinged with the weather on Friday," he said in an interview.

"The impact of the weather was certainly more than I had predicted."

"Mama" features Jessica Chastain as a woman forced to take care of two orphaned nieces who have been living in the woods. The $15 million production has now earned $48.6 million at U.S. and Canadian theaters through two weekends.

Chastain also stars in "Zero Dark Thirty" in an Oscar-nominated role as a dogged CIA agent searching for al Qaeda leader Osama bin Laden. The best picture nominee, which has sparked debate over depictions of torture, has grossed $69.9 million since its release in late December.

"Silver Linings Playbook," another Oscar contender, stars Bradley Cooper as a former mental patient trying to rebuild his life with the help of a young widow played by Jennifer Lawrence. Total sales for "Silver Linings" reached $69.46 million.

New crime thriller "Parker" finished in fifth place, taking in $7 million at domestic theaters. The film is based on a series of novels by Donald E. Westlake and stars Jason Statham as a thief seeking revenge against a crew that double-crossed him. Jennifer Lopez plays a woman who helps with his mission.

"Movie 43," a raunchy new comedy with an ensemble of Hollywood directors and stars, settled for seventh place. The film is a series of related short films about three kids scouring the Internet to find the world's most-banned movie. Stars making appearances include Huge Jackman, Seth MacFarlane, Kate Winslet and Dennis Quaid. Privately held Relativity Media produced the film for about $6 million.

"Mama" was distributed by Universal Studios, a division of Comcast Corp. Sony Corp's movie studio released "Zero Dark Thirty." "Parker" was released by independent studio FilmDistrict. The Weinstein Co distributed "Silver Linings Playbook."

Iranian workers demand South Korea 'return' funds

TEHRAN, Iran (AP) ? About 1,000 Iranian workers have staged a protest rally in front of the South Korean Embassy in Tehran demanding the return of $70 million allegedly held by Seoul after a collapsed business deal.

Employees of Iran's Entekhab Industrial Group claim South Korea has blocked a refund of the money after a failed bid in 2010 to buy Daewoo Electronics, a former subsidiary of the now-defunct Daewoo Group.

Iranians believe the deal was called off because of South Korean worries over mounting international sanctions against Iran over its nuclear program.

"Return our money," the protesters shouted Sunday in the third such rally in recent months.

Daewoo officials were not immediately available for comment.

Entekhab produces home appliances in Isfahan, about 300 kilometers (180 miles) south of Tehran.

Beginner Fly Fishing- Using The Proper Techniques And Practices ...

Before 200 AD, fly sportfishing was principally employed to capture angle such as trout and salmon. Many, who haven?t genuinely engaged in this sport will reckon the activity as uncomplicated. However, beginner fly fishing is not as trim as it seems ? it is not merely a simple task of connecting the rod and the line, splashing it in water for a bit and waiting for a catch. It requires practice; practice in proper fly fishing stratagies

The history of fly sportfishing runs back to the Macedonians, who after observing the Astraus River fish flying over the waters were able to catch fish using Hipporous flies. They exploited these flies to catch fish for food. That was in all likelihood how the name grew.

Today, fly fishing is practiced widely as a commercial fishing method and as a hobby. A wide form of fish like pike, panfish, carp and grayling, plus some saltwater varieties like snooke and bonefish are caught using fly angling techniques.

Nowadays, anglers make use of synthetic flies that are coated with plastic weighty enough to cast the rod at the directed spot without trouble. Some anglers, especially those who fly fish for fun, with the intention of releasing the catch afterwards, use all kinds of other natural material such as food leftovers, worms, etc.

In fly fishing, a line is cast rather than? a weighted lure. The fly line is lighter than a net and cannot be casted like one either. The physics of a fly cast is easy to picture. Once set in water, the object will produce an impulse or a wave which hits the senses of fish nearby. If you shake the cast a bit, ripples will be created in the water making fish to believe that there?s a treat for them.

There are unique? types of fly casts. A person must select a suitable cast matching to the atmosphere of the angling place and the purpose of the task.? These are some of the fly fishing methods. Sprey fishing uses heavier, double handled rods and is practiced to catch fish in more substantial reservoirs.

Fly fishing for trout is a popular sport. It is a fun procedure for recreation that allows you to practice not only skill, but also patience, energy and stamina. That is why fly fishin is highly recommended for people of all ages to be practiced as a sport.

PPC Campaign Strategies To Enhance Your Business's ...

PPC Campaign Strategies To Enhance Your Business?s Competetiveness, Visibility image paid search strategyJust like everything else, it is important to stay on top of the latest trends to make your business successful. When it comes to pay per click campaigns, several trends should not be ignore, to ensure that your business stays at the top. Whether or not you are just testing or considering a pay per click campaign, it is important to understand that you can take advantage of this very easily, based upon your specific needs and resources. You can easily evaluate your particular situation, based upon your web analytics. If you find expansion potential, consider implementing a PPC campaign that uses mobile extensions, mobile device conversions, display banners and more. If your e-commerce conversion rate is very low, you can make this a primary focus in your mobile PPC strategy, to maximize your ROI.

Micro-conversions are also on the horizon as the next great thing. It is important to note that these are not your primary pages. These are downloads of brochures, specific web pages, location checks, registering for newsletters, reading blog comments and more. These can add an exceptional amount of value to your business, because most people will not take action with one click. By evaluating your website and your PPC ads, you can discover the value of each click, including micro-conversions and other actions that you can take to encourage more engagement with your customers. You can also enhance your search engine ranking by taking advantage of video ad platforms. Whether YouTube or another video website, this is an exciting and valuable advertising portal. You don?t have to worry about making a viral video, all you need to do is consider your target audience, and create an innovative and relive video, including tips on how to use your products, technical support and much more, which can make them relevant and useful to your customers.

Moreover, although it may not be for everyone, you may be able to take advantage of dynamic generated ads. These give you the opportunity to check out creative and innovative ideas and market them to specific websites or users. Although this is very limited, if you are a display advertiser, this is extremely vital. This will give you the option of using Google?s Display Ad Builder and Merchant Center, and use their templates as well as products from their merchant feed. These will match up all relevant searches via the Internet directly to your website, provided you have a very specific campaign or structure of your keywords. It is also important to have a number of template ads that work cross product, as well as landing pages that match. Dynamic display ads are the next wave, so it is important to make sure that you keep up to date on this technology, take advantage of it and ensure your competitiveness in an ever-changing marketplace. Finally, although shopping has changed from traditional retail to more online, searching still plays a vital role. For this reason, you want to make sure that your PPC campaigns implement coupons, local searches, reviews and more and make sure that you can easily be found where your customers are looking for you.

IRS, Lockheed fight over R&D "rocket science" tax break

WASHINGTON (Reuters) - Lockheed Martin Corp is challenging the U.S. government in court over $13.6 million in research tax credits in a case that tests the often hazy line between research and production, with future R&D claims by other companies possibly at stake.

The aerospace group argued in December in U.S. District Court that the Internal Revenue Service had wrongly rejected research tax credits Lockheed claimed for two projects: a space rocket launcher and a New York City surveillance system.

The IRS disallowed Lockheed's claims in May 2012. Lockheed has challenged the IRS' position that some retroactive R&D credit claims were impermissible because they involved costs not for research, but for making prototypes resulting from research.

Lockheed said in its court filing that some of the credits were claimed for prototypes, but argued that the designs were new and unproven and should qualify as research.

"With Lockheed, you have the actual question of whether this is rocket science," said William Schmalzl, a partner at the law firm Mayer Brown with clients facing similar issues.

The case also draws attention to the timing of R&D credit claims. Lockheed's 2012 claims on the rocket launcher and surveillance system projects were both made retroactively for tax years 2004 through 2007, according to its court filing.

While retroactive claims are frequently allowed, they tend to attract closer IRS scrutiny, tax lawyers said.

When a credit is sought retroactively, "you really aren't relying on it as an incentive" to do research, Schmalzl said.

Maryland-based Lockheed, a top Defense Department supplier, declined to comment. The IRS also declined to comment.

The agency has yet to file a formal response to the Lockheed court action. No court date has been set in the U.S. District Court for the District of Maryland in Greenbelt.

A Lockheed court loss could jeopardize other companies' R&D claims and allow the IRS to be more aggressive, while a Lockheed victory could prompt other companies to look at past tax returns and claim retroactive credits, tax experts said.

PROMOTING SCIENCE

The R&D credit was created by Congress in 1981 to encourage business investment in science.

The "traditional" R&D credit allows companies to cut their tax bills by 20 percent of qualified research spending based on a formula dating back to the mid-1980s. An "alternative simplified credit," established in 2007, allows a claim of 14 percent based on the three prior years of research spending.

The credit is not refundable for cash, except in limited circumstances, and there is no cap on the credits a company can claim. Unused credits can be carried back one year and carried forward 20 years. The credit is limited to research funds spent in the United States. Wages and supplies account for the bulk of spending, and capital expenses are not applicable.

In reporting its quarterly financial results on Thursday, Lockheed said separately from the case that it expects to receive $75 million in R&D credits for 2012 and 2013.

Though simple in concept, the R&D credit is a frequent source of messy tax disputes. Companies, including Dow Chemical Co , Trinity Industries and Bayer AG , are also fighting the IRS in court over research credits.

Dow lost a case in U.S. appeals court in September involving $8 million in credits. Dow unit Union Carbide wanted to apply the credits retroactively to cover costs for supplies, even though it ended up selling the finished goods.

The Dow decision gave the IRS "ammunition to attack" certain credit claims for supply costs, said Jeff Malo, a director with tax advisory firm WTP Advisors.

In December, Dow asked the Supreme Court to hear its case.

Germany's Bayer is facing off against the IRS over $170 million of disputed research credits spanning 17 years. Texas-based Trinity is appealing a $5 million tax credit dispute.

"The confusion and ambiguity as to what qualifies, and the fights that this ambiguity engenders, undermine the credit's intended incentive effect," said Alex Sadler, a partner at law firm Ivins, Phillips & Barker.

The R&D credit is widely backed by President Barack Obama and members of both political parties, many of whom want to make it a permanent part of the tax code. It is presently a "temporary" measure requiring regular extension.

It was renewed again this month, along with other tax breaks known collectively as the "tax extenders." The R&D credit, which will expire again at the end of this year, is estimated to cost U.S. taxpayers $14.3 billion over 10 years.

This month the non-partisan Congressional Budget Office urged Congress to consider changes to the credit "to reduce excessive revenue costs and economic inefficiencies."

The case is Lockheed Martin Corp v. United States of America, in the U.S. District Court for the District of Maryland Southern Division No. 3725.

New Synthetic Polymer Is First to Match Rigidity of DNA or Collagen

The stiff supergel mimics cell scaffolding such that a cold solution of it poured on to a wound could quickly form a gel barrier when it warmed to body temperature

Take one kilogram of polyisocyanide polymer. Sprinkle liberally across an Olympic swimming pool. Warm gently. Within minutes, your jelly is ready. Serves 25 million.

Alan Rowan, a materials chemist at Radboud University Nijmegen in the Netherlands, is describing the properties of a remarkable polymer developed in his lab and unveiled today in Nature. He has not actually run the swimming-pool experiment, but he sounds as if he would love to give it a try. When it comes to forming gels, he says excitedly, his polymer is ?probably the best in the world ? an order of magnitude better than anything else?.

But it offers much more than record-breaking dessert portions: it is the first synthetic polymer that can match the rigidity found in many biological polymers, says Margaret Gardel, a biophysicist at the University of Chicago, Illinois, who wrote a News and Views article to accompany the publication. ?Nearly all biopolymers, like DNA or collagen, have some inherent rigidity,? she explains; synthetic polymers, by contrast, tend to be extremely floppy.

Rowan?s polymer strands have a helical backbone with thousands of short peptides jutting out from the sides, each carrying long tails made of repeating carbon and oxygen chains. Nitrogen and hydrogen atoms in neighboring peptides bond to each other to give the backbone rigidity, and the carbon and oxygen tails readily grab water molecules, making the polymer extremely soluble.

The polymer has a helical backbone (red) and tails (blue) that link to neighbouring strands.

Strong structure
Once the polymer is dissolved, warming it causes the tails to squeeze water molecules away and form links with neighboring polymer strands. Above a certain temperature, the solution transforms into a gel in seconds as the strands self-assemble into bundles roughly 10 nanometers wide. As with the biopolymers in a living cell, or the fibers in a rope, the bundling stiffens the whole structure. ?The nanoscale mechanism is the same as at the macroscale,? says Rowan.

Researchers already knew that bundling was important in strengthening biopolymers. But Rowan?s team has measured the stiffness of individual strands and of the bundles, and has shown the relationship between the two. ?Now that we understand the principles, we can start making gels at even lower concentrations,? predicts Rowan.

The polymer?s ability to form a gel on heating is relatively unusual: the opposite is true for most gels. A gelatin solution, for example, must be cooled to form jelly. Rowan imagines that a cold solution of his polymer poured on to a wound to protect the tissue would quickly form a gel barrier when it warmed to body temperature; applying an ice pack could liquefy it when it was no longer needed. The researchers are now running tests on this concept, ?using a leg of pork that we put in the oven at 40 ?C?, says Rowan.

Apple's new CEO ripe for success ? Business Management Daily ...

In 2011, Tim Cook replaced the late Steve Jobs as CEO of Apple. Since then, the 52-year-old has gradually established himself as a leader in his own right.

Cook joined Apple in 1998 as a senior vice president. But after 13 years at the company, no one knew how the first-time CEO would handle the reins.

Within months of becoming CEO, Cook differentiated himself from Jobs in highly visible ways. Responding to em??ployees? requests, Cook initiated stock-buyback plans and charitable-giving programs. Jobs mostly rejected these activities.

More boldly, Cook announced major management moves in October 2012. For starters, Cook terminated an executive that he had personally hired just six months earlier.

In firing John Browett, who ran Apple?s retail operation, Cook demonstrated that he was secure enough to acknowledge his hiring mistake sooner rather than later. Browett reportedly launched a scheduling system that cut employees? hours, stoking ire among staffers at Apple?s famous stores.

Leadership experts often advise CEOs to cut their losses quickly once they realize they?ve made a poor hiring decision. That?s what Cook did.

In another personnel move, Cook pushed out a senior executive who re??portedly refused to sign a public apology letter over Apple?s defective maps software in the new iPhone 5. Even before this incident, however, Cook was allegedly concerned by the executive?s difficult personality.

In another impressive display of leadership, Cook accepted responsibility for the errors in the maps software. He could have shifted blame to the departing executive who ran the software team, but Cook chose to take the heat in public before arranging for the executive?s exit a few weeks later.

How To Create A Successful E-mail Marketing Campaign | Internet ...

Most people have no idea how to successfully grow their online business through e-mail marketing even though it is one of the most effective ways to generate sales and customer loyalty. However, if you familiarize yourself with some of the tips that the pros use, then there is no reason why you cannot keep your customers interested in your business through simple email marketing efforts.

This article offers some tips about how to create a successful e-mail marketing campaign.

Frequency

If you send e-mails too frequently they may be regarded as ?spam? and people may remove themselves from your list.

On the other hand, if you fail to send e-mail frequently enough your subscribers may feel as if they?ve wasted their time signing up to your list. Remember your goals and audience so you can pick a happy medium for the frequency of your mailings.

SMS

In addition to using e-mail, incorporate SMS marketing into your campaign. Have customers sign up for an SMS option and send them text messages when on the go. When these two methods are used together, you have a better chance of getting more customers to sign up for your product or service.

?Above The Fold?

Think ?above the fold? when designing your messages. In newspapers, anything above the page-fold features a lot more prominently than material lower down the page. If someone has to scroll down to see your call-to-action element, it is much less likely that they will ever see it, let alone click it.

Trial Offers

A great suggestion for following up with clients is to send out a follow-up email that includes a trial offer. Include a line on your message that tells them to check it out right now. The closing postscript can include that this is an excellent bargain that they should take full advantage of.

Provide Quality Content

Every e-mail message you send out should provide your customers with good quality content that contains useful tips or information. Think of the newsletter as an opportunity to demonstrate your expertise in your field. Your customers will trust you more when they see how much you know and will be more likely to buy products or services from you.

Add A Disclaimer

When you invite your prospects to provide their email address during the sign-up process, make sure that a clear disclaimer is visible with some information about your newsletter content and frequency so they know that they will be receiving.

Write A Compelling Subject Line

The subject is one of the most important parts of your e-mail. If you do not have a subject that immediately grabs the reader?s attention, it is likely that they will not bother to read the e-mail at all. The best written e-mails can be completely worthless without a compelling subject. Make sure that all of your subjects are great!

Use Pre-Headers

Make use of pre-headers in your marketing emails. Certain e-mail clients ? the more advanced web-based ones in particular ? display a short summary for each email in the inbox. This is the pre-header. It is easy to set up. Clients generate pre-headers from the first line of text in the message. Compose your emails accordingly!

Identify Your Target Audience

When you are setting up an e-mail marketing campaign, it is pertinent to know who your target audience is. The proper research regarding your market niche will help you determine your target audience, and you can then build an effective email promoting campaign. Targeting the right customers is key to your success.

Making effective use of e-mail to promote your business can be tricky, but it gets easier as you get to know more about what you are trying to achieve. Done properly, e-mail marketing can dramatically boost your visibility and your sales.

Molecular switch enabling immune cells to better fight disease

Jan. 20, 2013 ? A research team led by the La Jolla Institute for Allergy & Immunology has discovered the mechanism that enables CD4 helper T cells to assume the more aggressive role of killer T cells in mounting an immune attack against viruses, cancerous tumors and other damaged or infected cells. The finding, made in collaboration with researchers from the RIKEN Institute in Japan, could enable the development of more potent drugs for AIDS, cancer and many other diseases based on using this mechanism to trigger larger armies of killer T cells against infected or damaged cells.

CD4 helper T cells, which normally assist other cells of the immune system during an infection, and CD8 killer T cells, which directly attack and eliminate infected cells, are two of the body's most important immune cells for defending against diseases. Earlier research studies have shown that helper T cells can become killer cells in some instances. However, the specific mechanism of action that allowed this to occur was not known until now.

"We have identified the molecular switch that enables CD4 T cells to override their programming as helper cells and transform into cytolytic (killer) cells," said La Jolla Institute scientist and study co-leader Hilde Cheroutre, Ph.D. "Our team also showed that these transformed helper T cells represent a separate and distinct population of cells. They are not a subset of TH-1 helper cells as previously thought."

Jay A. Berzofsky, M.D., Ph.D., chief of the Vaccine Branch at the National Cancer Institute's Center for Cancer Research, called the finding "a major advance" that provides new understanding about the cell's lineage and basic mechanisms. Dr. Berzofsky was among the researchers whose work in the 1980s first demonstrated that helper cells could convert to killer cells. "Understanding how these cells derive and what causes them to switch from helper T cells to cytolytic T cells is an important step to learning how to manipulate them in disease," he said, noting it could lead to novel approaches "either to turn these cells off in autoimmune disease or turn them on in infectious diseases."

He added that the finding could also have important implications in cancer. "We need all of the cytolytic machinery that we can get to try to destroy cancers," he said. "If we can learn to turn them on, I think it's reasonable to believe that these cytolytic T cells can play an important role in controlling cancer."

The findings were published January 20 in Nature Immunology in a paper entitled "Transcriptional reprogramming of mature CD4 helper T cells generates distinct MHC class II-restricted cytotoxic T lymphocytes." Dr. Cheroutre is co-senior author on the study together with Dr. Ichiro Taniuchi of the RIKEN Research Center for Allergy and Immunology in Yokohama, Kanagawa, Japan. First authors on the paper are: Mohammad Mushtaq Husain, Ph.D., of the La Jolla Institute; Daniel Mucida, Ph.D., formerly of the La Jolla Institute, now at Rockefeller University; Femke van Wijk, Ph.D., formerly of the La Jolla Institute, now at the University Medical Center Utrecht, The Netherlands, and Sawako Muroi, of the RIKEN Institute.

In the study, the researchers found that a certain transcription factor, which are molecules in the cell nucleus that control the activity of cells, continually suppresses the killer T cell lineage in helper T cells. Using mice, the team showed that turning off this transcription factor (ThPOK) enabled the helper cells in the body's peripheral areas, like the blood, spleen and the intestine, to override their original programming and to become killer T cells. "While our work focused on the intestines, we found that helper T cells in all tissues of the body have the potential to become killer cells in response to recognition of viral, tumor or other antigens in the context of cytokines such as IL-15," said Dr. Cheroutre.

Jonathan Braun, M.D., chair of the Department of Pathology and Laboratory Medicine at UCLA's David Geffen School of Medicine, praised the study as laying the groundwork for using T helper cells in a much more aggressive manner. "Helper T cells are mainly understood for their role in regulating other immune cells," he said. "This work reveals how they themselves can be triggered to become the action cells in the immune response. This opens new possibilities for how to manipulate them therapeutically in disease."

Dr. Cheroutre said the transformation of CD 4 helper T cells into killer cells already occurs in the body naturally. "Our finding could help to explain a number of occurrences that we haven't really understood up to this point, such as why some people can be chronically infected with HIV without developing AIDS." In these instances, Dr. Cheroutre is convinced that CD4 helper T cells must be taking over the role of killer cells after the CD8 T cells become exhausted. "It's like the helper cells can come in as reinforcements to keep the virus under control. If we can develop ways to artificially trigger that process, we may be able to significantly help people with HIV and other chronic infections."

While scientists would want to trigger a larger army of virus-specific killer cells in the case of infections, the opposite would be true in inflammation-fueled autoimmune diseases, like rheumatoid arthritis or multiple sclerosis, said Dr. Cheroutre. "The CD4 T cells are the bad wolves in inflammatory diseases because they often trigger more pro-inflammatory cells which worsen these conditions," she said. "With this knowledge, we may be able to prevent that by coaxing the CD4 killer cells to become regulatory cells instead, which is another one of their potential functions. In regulatory mode, the CD4 T cells suppress the immune system. This suppression reduces inflammatory cells, which is what we want to do in autoimmune diseases."

However in cancer, the CD4 T cell's regulatory function becomes problematic because they inhibit the killer T cells from destroying cancerous cells. This is because of their built-in mechanism to keep T cells from attacking the body's own cells, said Dr. Cheroutre. "Cancer cells develop from our own cells and only look a little different from healthy cells," she explained. "The killer T cells can sense that they are different and decide to eliminate them. However, the CD4 regulatory T cells frequently suppress the killer T cells and prevent them from destroying the cancerous cells. This is often how cancer cells can escape the immune system's normal action of stamping out bad cells."

Dr. Cheroutre said she believes it may be possible, using the newly discovered mechanism, to turn the CD4 regulatory T cells into killer cells that would aid, rather than block, the immune system's attack on cancerous cells.

Digital age prompting closure of base theaters

(AP) ? Stacey Darling loves watching family movies at the Ellsworth Air Force Base theater in South Dakota because it's so much more affordable than taking her three children to the multiplex in nearby Rapid City.

Darling, whose husband is an airman, has been catching second-run films on base for about 2 1/2 years, and was there Saturday for the theater's last showing ? a screening of the animated movie "Hotel Transylvania." The movie theater is among 60 around the globe run by the Army and Air Force Exchange Service that is screening its last picture show amid the industry's conversion to digital projection.

"We always come out for the cartoons," said Darling, of Grand Forks, N.D. "We like the family movies."

Darling said she wishes she could go to the theater even more now that her husband, Senior Master Sgt. David Darling, has deployed to southwest Asia.

It's just not cost effective for the exchange service to invest the $120,000 per theater needed to convert from 35 millimeter film to the new format at the theaters that are being closed, said spokesman Judd Anstey. Sixty theaters will make the upgrade.

"At locations where customer attendance is decreased due to a preference for off-installation entertainment venues, a determination has been made that continued operation is no longer a viable option," Anstey said.

The exchange service runs department-store-style retail outlets, fast-food restaurants, barbers and similar services on military facilities.

Saturday's "Hotel Transylvania" screening at Ellsworth Air Force Base drew about 250 people, thanks in part to the theater forgoing the admission price for its farewell flick.

Teri Marino wishes Ellsworth patrons knew earlier about the closure so they could try to raise the cash needed to save the theater, which showed its first flick in 1969.

Marino, whose husband is a master sergeant living on base, brought her 9-year-old daughter to the final show and was enjoying the camaraderie between the mothers.

"When you're in the military, you're like family," said Marino, 37. "Especially when our husbands are deployed, the mothers ? us women ? stick together."

Stateside base theaters are limited to second-run movies, which typically have already spent six weeks in an off-base multiplex before. Ticket prices are lower, as is attendance. Base theaters have also been hurt by increased competition from video streaming services and DVD rental kiosks, Anstey said.

"The digital age has certainly made it easier to stay home and get content delivered directly to one's own living room," he said. "In addition, the theater business has become more competitive with multiplex and IMAX theaters in many locations adjacent to military installations.

Of the theaters making the conversion to the digital format, about 26 are in the U.S. and about 34 are overseas for a total cost of about $7.4 million, Anstey said.

Camp Zama in Japan, Spangdahlem Air Force Base in Germany, Camp Arifjan in Kuwait and Fort Hood in Texas are the first four to make the jump, which is expected to be completed by the end of 2013, Anstey said.

David Burnett, force support squadron commander for the 28th Bomb Wing at Ellsworth, said base officials are working to ensure other movie options, such as more DVD kiosks and an arrangement with Carmike Cinemas in Rapid City for a military discount.

He said the theater has been an important spot for on-base families, especially those living in the dorms.

"It gives them an outlet," Burnett said. "It gives them something to do."

Ron Reynolds, who was stationed on the base years ago, has served as the theater's projectionist for more than a decade.

He loads a reel on one of the twin projectors before briefly turning down the upstairs room's speaker to make sure the sound is at its best.

"Projectionist is one of the greatest part-time jobs you'll ever have," said Reynolds, 49. "Sadly, all good things must come to an end."

Reynolds' son, 29-year-old Tyler Murphy, remembers seeing "Titanic" at Ellsworth as a teenager.

"This is a little bit of a nostalgic type of moment," said Murphy, now a teacher at Sturgis Brown High School. "It's never going to be the same anymore."

(AP) ? It's a busy week for Alicia Keys.

The singer-songwriter is set to perform at three events during Barack Obama's presidential inauguration on Monday. She'll sing the National Anthem at the Super Bowl on Feb. 3. And meanwhile, she popped over to Park City, Utah, to debut her first film as executive producer and composer.

The 32-year-old entertainer is attending her first Sundance Film Festival to support "The Inevitable Defeat of Mister and Pete," which premiered Friday. Directed by George Tillman, Jr., the film tells the story of two young boys who survive the streets of Brooklyn on their own.

"It's great because I love being part of bringing stories that you wouldn't often hear to the world," she said. "The fact that it was in my hometown ? New York ? that felt really good. The most important thing for me is that it looked and was so authentic."

This was Keys' first experience creating a film score, and she found the process enlightening.

"I think it really expanded me because it's a beautiful collaborative process," she said. "Being able to collaborate with (director) George (Tillman, Jr.), and he has such a cool feeling about music and is specific about how it related to each scene, and that was really interesting. There were some pieces that came really naturally to me and others that I had to kind of think more of how does that feel, what should that feel like?"

While she's thrilled to experience her first Sundance festival, Keys confessed she's constantly thinking about her Super Bowl performance.

"I'm really excited about it, I can't even lie," she said with a smile. "I have to rehearse it totally, as if it's a brand new song, because it is actually a brand new song in the style that I'll deliver it. I'm actually rehearsing it like a maniac."

Video Game Crafts ?N Gear is 10!? weeks old. There is more talent shining through than ever in this week?s edition, with consoles Nintendo themselves should have made as limited editions, Shadow of the Colossus? Quadratus guarding your all important computer files and Isaac Clarke cosplay so realistic that you?d be forgiven for believing we just put Dead Space 3 screenshots in.

Custom Nintendo Consoles

DeviantArt user Zoki64 has made some unbelievably beautiful mods of various Nintendo consoles, based on Nintendo franchises. After check out those highlights, remember to view all the work on his page, and his YouTube channel to appreciate all the hard work in motion!

Shadow of the Colossus Hard Drive

Unless you are the highly courageous (or downright insane) hero Wander, you probably wouldn?t choose to go head to head with the colossi. Who better to guard your important files and/or rude images, then, than Quadratus himself?

At first glance, it looks nothing more than an incredibly detailed and amazing statue. Yeah, I?m sold already. But this is actually also a USB 500GB external drive. Complete with glowing eyes, crafter foxfoxwaltz built the frame using cardboard, plaster, spray paint and who knows what else to create this beauty. I can?t imagine the how much time and effort went into this one.

Dead Space Cosplay

If only 2013 hadn?t just started ? then claiming this to be cosplay of the year already would sound far more honourable. Cosplayers Kevin and?L?a collaborate to become Dead Space?s Isaac Clarke and Ellie Langford. I?m convinced that they really are those characters, and not just dressing up.

While the light and fire effects were later added with photoshop, all of the armour is completely real. Add in some stunning photography work and you have images that could fool anyone into believing that these are official promotional photos or stills from a live action Dead Space movie. Actually, Kevin?s suit is so convincing that he has already done some promotional work for EA!
